Abstract

The objective of the present work is to develop a colon specific drug delivery system by use of guar gum and chemically modified guargum in different proportions by employing ibuprofen as a model drug. The tablets were prepared by wet granulation method by use of PVP as binder. The release of drug from matrix tablets is influenced by polymer’s swelling behavior, gelling property and degradation by colonic bacteria. The prepared tablets were evaluated for hardness, weight variation, drug uniformity, friabilty and invitro drug release. The dissolution data revealed that the tablets containing guar gum and crosslinked guargum in higher concentrations each showed 83.246±0.32% and 75.125± 0.37% of drug release respectively within 24hrs study period. Results clearly indicate that decrease in drug release with increase in the polymer concentration. The results were subjected to study the release kinetics. The values of correlation coefficient indicate that drug release followed zero order release kinetics with Peppas drug release mechanism.
